At D.O.OTT, I have the privilege to develop, learn new skills and be a
part of the current research project: (Development of approaches to
spinal muscular atrophy therapy on the cellular model of the disease)
In this capacity, my tasks usually cover the following matters:
▪ Dealing with fibroblast cell cultures obtained from patients with
SMA (reagents, media, incubating, etc...)
▪ Fibroblast transfection with the combination of peptide carrier and
antisense oligonucleotide developed earlier in the laboratory
▪ RNA extraction and RNA concentration measuring using
NanoDrop spectrophotometer
▪ cDNA synthesis
▪ Polymerase Chain Reaction PCR
▪ Visualization and processing of PCR results using polyacrylamide
gel electrophoresis
At D.O.OTT, I also had the chance to be part of the project:
(Establishment of a Pilot Newborn Screening Program for Spinal
Muscular Atrophy in Saint Petersburg) which was incorporation with
Novartis Farma LLC.
At D.O.OTT, I also had the chance to be part of the project:
(Determination of the SMN1 gene copy number using LNAoligonucleotide probes). During that project my tasks covered the
following skills:
▪ DNA extraction and isolation from blood samples as well as from
dry blood samples
▪ Optimization of real-time PCR procedure with the new LNA
probes developed earlier in our laboratory
▪ Performing real-time PCR runs on samples for a cohort of
healthy individuals, SMA carriers and SMA patients as
negative controls and comparing these results with those
that already obtained with MGB probes
